仿刺参体壁水提物中生物大分子的鉴定

摘 要 采用匀浆水提处理获取仿刺参体壁水提物,测得其中蛋白质质量分数为(41.03±1.97)%,多糖质量分数为(36.51±1.58)%。基于蛋白组学技术鉴定出水提物中蛋白分子400余种,丰度较高的蛋白主要是卵黄蛋白、原肌球蛋白及肌动蛋白等。单糖组成分析和核磁共振波谱分析确认水提物中的多糖主要为岩藻聚糖硫酸酯,其分子质量为(5 172.5±107.6) kDa;利用多种变性剂破坏水提物中可能存在的分子间相互作用,发现处理前后岩藻聚糖硫酸酯分子量无显著变化,说明该多糖以非聚集体的形式存在于水提物中。该研究首次明确了仿刺参体壁水提物中的蛋白质和多糖的分子种类,可为后续优化海参加工工艺、减少海参加工中的营养物质流失提供理论指导。

海参是棘皮动物门海参纲海洋动物的总称,是我国传统海产珍品,现已证实海参体内含多种生物活性物质,其营养价值和滋补功能受到消费者青睐[1-2]。仿刺参(Apostichopus japonicus)是我国主要养殖海参品种,具有重要的经济价值,据统计,2018年我国仿刺参养殖量达17.4万t[3]

仿刺参容易发生自溶,不易于运输及贮藏,多以海参加工制品的形式流通。在海参加工过程中,需经过预煮、浸泡、煮制及(或)泡发等环节,在此过程中海参体壁中的营养成分会发生不同程度的流失。现有研究表明海参加工过程中随水流失的营养成分主要是蛋白质及多糖类,如QI等[4]对海参加工废液中的成分进行了测定,结果表明海参加工废液固形物中的多糖质量分数为(69.69±4.52)%、蛋白质质量分数为(14.06±2.31)%、灰分质量分数为(12.25±1.28)%、脂肪质量分数为(1.19±0.64)%;孙昊等[5]报道,海参加工水煮液中的总糖、蛋白质、灰分及脂肪分别占干重的48.50%、35.25%、12.25%及1.19%。陈宁等[6]对仿刺参水煮液中糖蛋白的单糖组成及氨基酸组成进行了分析,结果表明鲜活仿刺参经水煮、醇沉及脱盐处理后得到的样品中含有氨基葡萄糖、甘露糖、氨基半乳糖、葡萄糖、半乳糖、岩藻糖等单糖及18种氨基酸。然而,对于随水流失的蛋白质及多糖的分子种类目前尚缺乏系统报道。

本研究以匀浆水提的方式处理仿刺参体壁,以充分释放海参体壁中可能被水抽提的成分,获得仿刺参体壁水提物,在此基础上利用蛋白组学技术研究水提物中蛋白质的分子组成,以单糖组成分析及核磁共振波谱技术(nuclear magnetic resonance,NMR)确定多糖种类,以期为后续海参加工工艺的优化、减少海参加工中的营养物质流失提供理论指导。

1 材料与方法

1.1 试剂与设备

鲜活仿刺参(约300 g/只,参龄3年),青岛市市南区水产品市场。

乙腈(色谱纯),德国Merck公司;次甲基蓝(分析纯),上海试剂三厂;三氟乙酸(trifluoroacetic acid,TFA),分析纯,上海化学试剂公司;重水(D2O),分析纯,Fluka公司;三甲基硅丙磺酸钠(2,2-Dimethyl-2-silapentane-5-sulfonate, DSS),分析纯,美国Sigma公司; 1-苯基-3-甲基-5-吡唑啉酮(1-phenyl-3-methyl-5-pyrazolone, PMP),分析纯,上海麦克林生化科技有限公司;甲醇、乙醇、二硫苏糖醇、NaH2PO4、Na2HPO4、HCl、NaCl、乙酸铵、H2SO4、苯酚、NaOH、尿素、Na2CO3、叠氮钠、乙二胺四乙酸(ethylene diamine tetra acetic acid, EDTA)等试剂, 均为国产分析纯,国药集团化学试剂有限公司;木瓜蛋白酶(分析纯),上海源叶生物科技有限公司;胰蛋白酶(质谱纯),美国Thermo Fisher Scientific公司;蛋白酶抑制剂(分析纯),北京天恩泽基因科技有限公司。

Agilent 1260高效液相色谱仪、示差检测器、紫外检测器、ZORBAX Eclipse XDB-C18色谱柱(4.6 × 250 mm), 美国Agilent公司;KTA prime plus蛋白纯化系统、HitrapTM Desalting脱盐柱、HiPrep 16/60 Sephacryl S-500 HR,美国GE公司;Dawn Heleos-II多角度激光光散射仪,美国Wyatt Technology公司;TGL-16C高速冷冻离心机,上海安亭科学仪器厂;Bruker AVANCE 600 MHz核磁共振仪,德国Bruker公司;OHpak SB-807 HQ(8.0 mm × 300 mm),日本Shodex公司;Acclaim PepMap RSL C18分析柱(75 μm×150 mm,2 μm),美国Thermo Fisher Scientific公司;InertSep RP-C18固相萃取小柱(5010-27130),北京谱朋科技有限公司。

1.2 仿刺参体壁水提物的制备

对鲜活仿刺参进行解剖,取体壁组织并切块,加入5倍组织块鲜重的超纯水并加入蛋白酶抑制剂,匀浆后置于磁力搅拌器上,于4 ℃搅拌12 h,以15 000×g离心30 min取上清液。上清液冻干,得到仿刺参体壁水提物。

1.3 仿刺参体壁水提物的基本成分分析

采用福林酚法[7]、硫酸-苯酚法[8]对仿刺参体壁水提物中的蛋白、多糖含量进行测定。按照美国分析化学家协会 (Association of Official Analytical Chemists, AOAC)[9]方法对仿刺参体壁水提物中的脂肪,水分与无机盐含量进行测定,以上述成分相对于水提物干重的质量分数表征其含量。

1.4 仿刺参体壁水提物的蛋白组分析

1.4.1 前处理

将仿刺参体壁水提物溶解于超纯水中,在4 ℃下12 000×g离心10 min,取上清液即得样品总蛋白。按照WISNIEWSKI等[10]的方法对样品进行胰蛋白酶酶解,酶解后的肽段采用C18固相萃取柱脱盐,上样后以0.1% TFA水溶液冲洗3次、体积分数90%乙腈-水(含体积分数0.1% TFA)洗脱3次,洗脱液真空抽干后以体积分数0.1%甲酸-水复溶。

1.4.2 液质分析

按照FIRMINO等[11]的方法,采用C18分析柱对样品进行分离,流速为300 nL/min,梯度洗脱90 min。所有MS/MS图谱采集使用数据依赖型的正离子模式下的高能碰撞裂解完成,全扫描在300~1 600 m/z进行,分辨率设为17 500,最大注射时间为80 ms。

1.4.3 数据库检索

参考COX等[12]的方法利用MaxQuant(版本号1.3.0.5)进行搜库,并进行蛋白丰度非标定量分析。搜库参数设置如下:数据库,美国国家生物技术信息中心(US National Center for Biotechnology Information, NCBI)仿刺参蛋白质组;假阳性率≤0.01;固定修饰,氨基甲酰甲基(C);可变修饰,氧化(M);酶,胰蛋白酶;MS/MS容差,0.5 Da;MS容差,20 ppm。

1.5 仿刺参体壁水提物中多糖组分的分析

1.5.1 多糖组分的分离纯化

仿刺参体壁水提物以超纯水复溶后,以截留分子量为8 kDa的透析袋进行透析脱盐并冻干。冻干物溶解于50 mmol/L NaH2PO4-Na2HPO4(pH 7.0)缓冲液中,16 000×g离心30 min。利用Hiprep 16/60 Sephacryl S-500凝胶色谱柱分离纯化样品中的多糖组分,以0.2 mol/L NaCl洗脱,流速为0.5 mL/min。使用次甲基蓝法检测收集各管中的阴离子多糖[13],并绘制洗脱曲线。收集各管洗脱峰,并利用HitrapTM Desalting脱盐柱脱盐。

1.5.2 单糖组成分析

以PMP柱前衍生的高效液相色谱法测定样品的单糖组成[14]。配制质量浓度为2.364 mg/mL多糖水溶液,取0.846 mL于安瓿瓶中,加入0.154 mL 13 mol/L TFA,使多糖浓度为2 mg/mL、TFA浓度为2 mol/L。充氮气并封管,于110 ℃烘箱中水解10 h后冷却至室温,于80 ℃水浴中挥干TFA。加入200 μL超纯水冲洗溶解,用0.3 mol/L NaOH溶液调至pH中性,转移至1.5 mL离心管内,最终定容至1 mL。取450 μL定容后的样品加入50 μL乳糖内标溶液、450 μL 0.5 mol/L PMP溶液及450 μL 0.3 mol/L NaOH溶液,70 ℃水浴反应30 min完成衍生化。加入450 μL 0.3 mol/L的HCl进行中和,并用氯仿萃取除去PMP,得到的水溶液以液相色谱进行分析。液相色谱条件为:ZORBAX Eclipse XDB-C18分离柱;柱温25 ℃;紫外检测器;流动相,A为0.1 mol/L NH4Ac-HAc缓冲液(pH 5.5),B为乙腈;流速1.0 mL/min;体积梯度,45%~100% B;时间梯度,0~45 min。

1.5.3 NMR分析

利用配备有超低温探头的600 MHz核磁共振仪对收集的多糖进行1H核磁共振分析[14],取50 mg样品,经D2O(99.9%)连续置换3次,置换结束后用0.5 mL D2O(99.9%)复溶,置于核磁管中进行1H核磁共振测定,测试温度为25 ℃,以DSS作为外标。根据YU等[2]的方法制备仿刺参岩藻聚糖硫酸酯,作为参考样品进行比较分析。

1.5.4 分子质量分析

以高效凝胶排阻色谱-多角度激光光散射-示差折光检测法(high performance size exclusion chromatography-multiangle laser light scattering-refractive index,HPSEC-MALLS-RI)[15]测定多糖分子质量。液相色谱条件为:OHpak SB-807 HQ;柱温40 ℃;流动相:0.15 mol/L NaCl (含有10 mmol/L NaH2PO4-Na2HPO4缓冲液,pH 7.4);流速0.5 mL/min;进样体积20 μL;dn/dc(0.1147±0.0012) mL/g[15]

1.5.5 多糖与水提物中其他生物大分子的相互作用分析

用0.1 mol/L NaOH、木瓜蛋白酶、2 mol/L NaCl或 8 mol/L尿素(含10 mmol/L EDTA)破坏水提物中多糖与其他生物大分子之间可能存在的相互作用。水提物加入质量分数3%的木瓜蛋白酶,在60 ℃水浴下反应12 h;其他变性剂的处理在4 ℃下进行12 h。对处理后的样品离心(NaOH处理后的样品离心前先进行中和),收集上清液,经HiPrep 16/60 Sephacryl S-500色谱柱分离并绘制洗脱曲线,方法如1.5.1所述;对于NaCl或尿素处理后的样品,分离流动相分别为2 mol/L NaCl及8 mol/L尿素(含10 mmol/L EDTA)溶液,以防止分子间相互作用的重新形成。

1.6 统计分析

各实验重复3次,结果以平均值±标准偏差表示。显著性差异以SPSS 16.0软件进行分析,以P<0.05作为显著性差异的判断标准。

2 结果与分析

2.1 仿刺参体壁水提物中的基本组成

仿刺参体壁匀浆后得到的水提物相较仿刺参组织块干重的得率为(47.10±1.06)%,水提物中蛋白质、多糖、无机盐、水分、脂肪质量分数分别为(41.03±1.97)%、(36.50±1.58)%、(18.31±1.21)%、(2.41±0.31)%、(1.75±0.19)%。水提液中的营养成分主要以蛋白质及多糖为主,这与前人关于海参水煮液等的研究结论相符[4-6],但具体数值有一定差异,可能由于提取方法不同导致。

2.2 仿刺参体壁水提物中的非标记定量蛋白质组学分析

利用非标记定量蛋白质组学技术从仿刺参体壁水提物中共鉴定出蛋白400多种(附件1),说明仿刺参体壁水提物中蛋白质种类丰富。蛋白质组学鉴定丰度较高的蛋白见表1,结果表明卵黄蛋白类的丰度最高,还存在一定量的肌肉蛋白组分包括原肌球蛋白、肌动蛋白、肌球蛋白等。

FUJIWARA等[16]的研究结果表明仿刺参的体腔液中含有卵黄蛋白;LI等[17]的研究表明卵黄蛋白在海参成体组织(肠、纵肌、性腺、体壁等)中有不同的表达。卵黄蛋白除了具有丰富的营养价值,其水解产物多肽还具有多种免疫活性,DENISE等[18]研究表明卵黄蛋白肽具有抗氧化活性。卵黄蛋白原可破坏细菌的细胞壁,其杀菌及抗氧化功能已被广泛报道[19];SUN等[20]指出卵黄蛋白原对人体健康具有潜在功效。仿刺参体壁水提物中的多种蛋白均具有良好的生物活性与免疫功能,海参加工中随水流失的物质可作为制备活性蛋白及活性多肽的原料。

仿刺参体壁水提物中含有一定量的原肌球蛋白及肌动蛋白,这些蛋白是组成肌肉蛋白的主要成分,属于盐溶性蛋白[21]。海参属于海洋生物,据报道,海参体壁盐浓度与海水盐浓度一致[22],仿刺参体壁水提物中盐的质量分数较高,为(18.31±1.21)%,预示海参匀浆后水提液中的盐浓度较高,从而导致盐溶性蛋白的溶出。

仿刺参体壁水提物中也发现了胶原蛋白组分。胶原蛋白属于水不溶性蛋白,是海参体壁中含量最丰富的生物大分子成分[23]。海参体内含有丰富的自溶酶,导致海参易于自溶[4],虽然本研究在海参体壁匀浆水提的过程使用了蛋白酶抑制剂,但仍无法避免部分胶原蛋白被降解,因此推测少量胶原蛋白的出现可能是由于在匀浆过程中海参自溶酶降解胶原蛋白所致。

2.3 仿刺参体壁水提物中多糖组分的分析

如图1所示,仿刺参体壁水提物中的糖类物质经过HiPrep 16/60 Sephacryl S-500色谱柱分离纯化后,以次甲基蓝显色,表现为1个多糖峰。已有研究表明,仿刺参体壁中存在2种多糖类物质,即岩藻聚糖硫酸酯及硫酸软骨素,两者均属于酸性多糖[24]。收集凝胶色谱洗脱的多糖组分,并对该组分进行单糖组成测定,结果表明,岩藻糖是该多糖的优势单糖,占所有组成单糖质量分数的(96.87±0.03)%。据先前研究报道,仿刺参岩藻聚糖硫酸酯由岩藻糖及组成[25]。水提物中多糖组分的单糖组成与仿刺参岩藻聚糖硫酸酯的单糖组成特征吻合。

本实验室前期已解析出仿刺参岩藻聚糖硫酸酯的结构,仿刺参岩藻聚糖硫酸酯的核磁共振1H谱图已经明确[2]。采用NMR对水提物多糖组分与仿刺参岩藻聚糖硫酸酯进行比较分析,结果表明,水提物多糖组分的1H NMR谱图与仿刺参岩藻聚糖硫酸酯的1H NMR谱图吻合(图2)。上述结果表明水提物中的主要多糖组分为岩藻聚糖硫酸酯。仿刺参体壁中的岩藻聚糖硫酸酯可以通过水提获取,说明岩藻聚糖硫酸酯分散于仿刺参体壁组织的细胞外基质中。最近的研究表明,仿刺参体壁中的硫酸软骨素以O-糖苷键的形式共价结合于体壁胶原纤维[26],仿刺参体壁水提物中并未检测到大量的硫酸软骨素组分,与该报道结论一致。

利用HPSEC-MALLS-RI测得仿刺参体壁水提物中的岩藻聚糖硫酸酯的分子质量为(5 172.5±107.6)kDa。YU等[2]提取得到的仿刺参岩藻聚糖硫酸酯的分子质量为1 970 kDa;LIU等[27]从仿刺参体壁提取出的岩藻聚糖硫酸酯的分子量为36.2 kDa;KARIYA等[28]通过氯仿-甲醇法从日本刺参(Stichopus japonicus)体壁中分离出2种岩藻聚糖硫酸酯,分子质量分别为9 kDa与32 kDa。本研究中测得的岩藻聚糖硫酸酯的分子量高于先前报道,为进一步探究水提物中的岩藻聚糖硫酸酯是否以聚集状态存在,使用多种变性剂或酶处理仿刺参体壁水提物,破坏其中生物大分子间可能存在的相互作用:以尿素破坏氢键相互作用;以EDTA破坏分子间盐桥;以NaCl屏蔽静电相互作用;以NaOH通过β-消除反应裂解碳水化合物与蛋白质之间的O-糖苷键;以木瓜蛋白酶破坏肽键[25]。结果表明,处理前后仿刺参体壁水提物中的岩藻聚糖硫酸酯在HiPrep 16/60 Sephacryl S-500色谱柱上出峰时间没有发生变化,说明仿刺参体壁水提物中的岩藻聚糖硫酸酯不存在明显的聚集,即以非聚集体的形式存在。不同研究提取出的岩藻聚糖硫酸酯分子量大小存在差异的原因可能是由于提取方法和过程不同导致。本研究对鲜海参进行匀浆水提,提取过程流程短,获得的岩藻聚糖硫酸酯更接近鲜活海参体壁中的状态。在生物体中,高分子量多糖并不罕见,比如透明质酸的平均分质量约为6 000 kDa[29]。本研究结果显示,仿刺参体壁中的岩藻聚糖硫酸酯分子质量大,推测岩藻聚糖硫酸酯可能在体壁中作为填充物,对仿刺参体壁空间结构的维持发挥着重要作用。

3 结论

本文研究了仿刺参体壁匀浆水提物中的蛋白质与多糖的分子组成。利用非标记定量蛋白质组学技术检测出蛋白质分子400多种,丰度较高的蛋白主要为卵黄蛋白、原肌球蛋白及肌动蛋白等。通过单糖组成分析和NMR技术确定水提物中的多糖主要为岩藻聚糖硫酸酯,分子质量为(5 172.5±107.6)kDa;利用多种变性剂破坏生物大分子间可能存在的相互作用,处理前后多糖分子量未发生显著变化,说明岩藻聚糖硫酸酯以非聚集体的形式存在。仿刺参体壁水提物中生物大分子分子组成的确定将为优化海参加工工艺、减少海参加工中营养成分的流失提供理论指导。

Identification of biomacromolecules in water extract of the body wall of sea cucumber Apostichopus japonicus

ABSTRACT Sea cucumber is an important economic aquatic product in China. In this study, water extract in the body wall of Apostichopus japonicus was obtained by homogenization and water extraction. The protein and polysaccharide content in the water extract was(41.03 ± 1.97)% and (36.50 ± 1.58)%, respectively. Proteomics revealed that the water extract contained more than 400 types of protein molecules, majorly including yolk protein, tropomyosin and actin. Monosaccharide composition analysis and nuclear magnetic resonance spectroscopy analysis confirmed that the polysaccharide in the water extract was mainly sulfated fucan (FUC) with a molecular weight of (5 172.5 ± 107.6) kDa; various denaturants were used to destroy possible molecules in the water extract between them. It was found that the molecular weight of FUC did not change significantly before and after treatment, suggesting that FUC existed in a non-aggregated form. This study identifies for the first time the molecular types of proteins and polysaccharides in the water extract of the sea cucumber body wall, which could provide theoretical guidance for the subsequent optimization of sea cucumber processing techniques and reduction of the nutrient loss during the processing.
