Abstract

To enhance the productivity of proline from Corynebacterium acetoacidophilum(originated from glutamic acid producing strain,ATCC-13870),a novel mutation system with atmosphere and room temperature plasmas(ARTP) was used.Under a cell lethality rate of 99%,accompanying with a high throughput screening method using 48-well microplates,16 mutants with changes on growth rates and proline production as compared with wild type were obtained.Experiment results indicated that the proline concentration produced by D3 mutant was increased from 5.47(g/100 mL) of the wild strain to 6.58(g/100 mL) after 48h fermentation.
