Medium optimization for Chlorella sp. MBFJNU-17 heterotrophic cultivation

Abstract

Urea was selected as the single nitrogen source for Chlorella sp. MBFJNU-17 heterotrophic cultivation. The results showed that the maximum biomass and the protein content of Chlorella was 10.85 g/L and 44.5% in batch cultivation using urea as nitrogen source. Meantime, the stable pH of cultures could benefit for the growth of Chlorella treated by urea. The significant factors that affected the growth of Chlorella were screened via Plackett-Burman experiments. Further, the concentrations of glucose, urea and trace elements stock solution were optimized by response surface methodology. And the biomass of Chlorella cultivated in the optimized medium was significantly increased to 15.53 g/L with the protein content of 52.3%. The essential amino acid index (EAAI) of Chlorella sp. MBFJNU-17 protein and soybean protein were compared with FAO/WHO/UNU standard. The results showed that the EAAI of Chlorella was 0.724 which higher than soybean protein (0.657). Taken together, Chlorella sp. MBFJNU-17 attained high density of biomass by the medium optimization, and exhibited more nutritional value than soybean protein, which can develop for animal feed or human food protein.
