Screening of fucoidanase-producing strains and preparation of low molecular weight fucoidan by enzymatic hydrolysis

Abstract

In this study, a new fucoidanase-producing strain was screened from Daqu by using fucoidan as the only carbon source with enzyme activity detection, which was identified as Aspergillus chevalier by morphology and molecular biology and the fucoidanase activity of this fungi was 0.414 U/mL. The low molecular weight fucoidan (<10 kDa) was prepared by hydrolysis with fucoidanase, and the enzymatic hydrolysis conditions were optimized by single factor and orthogonal experiments. The results showed that under the conditions of substrate concentration of 15 g/L, pH 6.0, the enzymatic hydrolysis temperature of 55 °C, and the enzymatic solution time of 24 h, the highest yield of low molecular weight fucoidan was up to 52.85%. The low molecular weight fucoidan obtained by enzymatic hydrolysis was purified in a graded manner, and three products with different molecules were obtained: F1 was 1 250 Da, F2 was 2 505 Da, and F3 was 8 227 Da. The antioxidant results showed that three products have a good antioxidant activity than fucoidan, this indicated that it is feasible to prepare low molecular weight fucoidan with molecular weight less than 10 kDa by hydrolysis of fucoidanase from A. chevalieri WA1207, which lays the foundation for the subsequent analysis of fucoidan structure and investigation of the relationship between function and structure.
