The purpose of this paper was to establish an animal model targeting female constipation, which could be used to screen drugs or probiotics suitable for female constipation.Previous studies have found that hormones, especially estradiol and/or its receptors, play an important role in female constipation.Based on this, this paper mainly discussed the effect of hormone receptors, especially estradiol receptors, on constipation.The rats were given tamoxifen citrate tablets, an estrogen receptor β inhibitor, for 7 days to analyze the effect of estrogen receptor β downregulation on constipation, intestinal nerve density and intestinal barrier.The opioid receptor inhibitor loperamide, which was a traditional model drug for constipation, was used as a positive control drug to clarify the pathway of estrogen receptor β affecting constipation.Results showed that the loss of estrogen receptor β may result in damage to the colonic nerve and intestinal barrier.It mainly showed that the density of intestinal glial cells, goblet cells, and substance P decreased, and the abundance and structure of intestinal flora changed.
