Effect of three monoterpenoids on triterpenoids production of Antrodia cinnamomea in petri-dish culture

Abstract

Antrodia cinnamomea is a kind of edible medicinal fungus.Triterpenoids are one of the dominant active ingredients in the fruiting bodies of A. cinnamomea, which have pharmacological effects such as anti-cancer, hepatoprotective and anti-inflammatory.Wild fruiting bodies of A. cinnamomea are only found on the inner rotten wall of Cinnamomum kanehirae, which leads to the fact that the active products of A. cinnamomea cannot satisfy the huge market demand.Petri-dish culture is a method for artificial cultivation of A. cinnamomea, which has the advantages of short cultivation time and simple operation.But the triterpenoids production of the A. cinnamomea mycelia in petri-dish culture is low.C. kanehirae is the only wild host of A. cinnamomea.Previous studies had indicated that extracts of C. kanehirae could promote the growth and triterpenoids production of mycelia in petri-dish culture.However, there are few reports on the effects of a single substance from C. kanehirae on triterpenoids synthesis of A. cinnamomea in petri-dish culture.To enhance the tirterpenoids production of A. cinnamomea mycelia, three monoterpenoids (α-terpineol, terpinene-4-ol and α-terpinene) from C. kanehirae were added in the medium in petri-dish culture.Biomass and triterpenoids yield of the mycelia were used as detection indices.The results indicated that α-terpineol was the only one among the three monoterpenoids had the ability to promote the growth of A. cinnamomea mycelia in petri-dish culture.The optimal adding concentration of α-terpineol was 0.08 g/L and the biomass of A. cinnamomea mycelia reached to 165.67 mg/plate.Three monoterpenoids all have the promotion effect on synthesis of triterpenoids in A. cinnamomea in petri-dish culture.The optimal adding concentration of α-terpineol is 0.08 g/L and the triterpenoids yield of A. cinnamomea mycelia reached to 17.55 mg/g with 50.39% increase compared with the control group, in the meanwhile, one triterpenoid, dehydrosulphurenic acid, was detected 3.20-fold increase compared with the control group.The optimal adding concentration of terpinene-4-ol is 0.12 g/L and the triterpenoids yield of A. cinnamomea mycelia reached to 8.89 mg/g with 28.65% increase compared with the control group.The optimal adding concentration of α-terpinene is 0.16 g/L and the triterpenoids yield of A. cinnamomea mycelia reached to 13.56 mg/g with 55.68% increase compared with the control group, in the meanwhile, two triterpenoids, dehydrosulphurenic acid and dehydroeburicoic acid, were detected 5.56-fold and 0.96-fold increase respectively compared with the control group.In this study, three monoterpenoids that could promote triterpenoids production of A. cinnamomea in petri-dish culture were identified and it provides new ideas for promoting triterpenoids synthesis of A. cinnamomea in petri-dish culture.
