Abstract

Investigating the effects and mechanisms of Bifidobacterium breve CCFM1489 combined with Cassiaside in alleviating metabolic dysfunction-associated fatty liver disease(MAFLD).Five-week-old male C57BL/6 J mice were selected and randomly divided into control group, model group, atorvastatin group, Cassiaside group, CCFM1489 group, synbiotic group, and postbiotic group, and were fed with high-fat diet for 8 weeks to establish a MAFLD model.Following 4-week intragastric drug administration, this study assessed the obesity characteristics, glucose metabolism capacity, blood lipids, body inflammation, liver injury, and liver pathology of each group, combining with the 16S rRNA intestinal flora sequencing and untargeted metabolomics to reveal the mechanism of action of Bifidobacterium breve CCFM1489 in combination with Cassiaside to alleviate MAFLD.Compared with the model group, postbiotics significantly reduced body weight, liver weight ratio, area under the oral glucose tolerance test (OGTT) glucose curve, TC, TG, LDL-C, HDL-C, ALT, AST, and body inflammation in MAFLD mice.Hematoxylin-eosin staining and oil red O staining showed that the postbiotic group was able to significantly improve liver pathomorphology and reduce fat accumulation in the liver.Intestinal flora analysis showed that the intervention of postbiotic was able to increase the abundance of Bacteroidetes and decrease the abundance of Firmicutes at the phylum level, and specifically decrease the abundance of Erysipelatoclostridium, Ruminococcaceae UCG-004 at the genus level.Metabolomics analyses suggested that postbiotics may exert a mitigating effect on MAFLD by modulating glyoxylate and dicarboxylic acid metabolism, taurine and hypotaurine metabolism, and glutathione metabolism.Bifidobacterium breve CCFM1489-Cassiaside postbiotics alleviated the pathological process of MAFLD by regulating the intestinal flora structure and modulating the glyoxylate/dicarboxylic acid metabolism, taurine metabolism and glutathione metabolism, providing a new idea for the microecological intervention strategy of MAFLD.
