In this study,the culture medium element and fermentation conditions of Bifidobacterium bifidum were optimized to determine the technical conditions that are beneficial for industrial batch production at low cost.Four out of six factors,namely tomato juice,fructo-oligosaccharide,waste molasses,potato starch,corn steep liquor,bean cake powder,were selected through single factor experiments.By conducting orthogonal experiments with the orthogonal table L9(3)4,we determined the optimum medium formula and the favorite fermentation conditions.The medium formula were:tomato juice 80 ml/L,fructo-oligosaccharide15 g/L,waste molasses 20 ml/L,corn steep liquor 110 ml/L,0.1%resazurin1 ml/L,tween-80 1 ml/L,salt solution 40 ml/L,L-cysteine hydrochloride0.5 g/L;the favorable fermentation conditions were temperature 40 ℃,initial pH 6.8,inoculation amount 3%,and culture hour 32 h.The low production cost and the easy-to-control fermentation conditions make the optimum medium element suitable for industrial batch production of Bifidobacterium bifidum.
