Culture Conditions of Transgenic Volvariella volvacea for High Production of Muli-Functional Cellulase

Abstract

TVM186 with high MFC activity was separated by monospore isolation from the transformants of Volvariella volvacea.Solid substrate fermentation was performed to investigate the optimum fermentation conditions for enzyme production of the transgenic V.volvacea.Several single factors on fermentation of strain TVM186 were studied in this research.The result showed that the optimum culture conditions of solid state fermentation were wheat bran power and soybean power with allocated proportion is 1:1.The optimum solid state fermentation conditions of TVM186 were initial pH 7.4,inoculation 6%,temperature 34℃,fermentation time 10 d,water ratio 64%,substrate 18.5 g using uniform design experiment.The highest Xylanase and CMCase activity were 1 427.38 U/g and 97.59 U/g respectively.
