In this paper, the effect of glucose and glycerol was investigated on the production of docosahexaenoic acid (DHA) by Aurantiochytrium limacinum. It was found that using glucose as carbon sources the substrate consumption rate and the cell growth rate were significantly higher than those with glycerol as carbon source, meanwhile using glycerol as carbon source the proportion of DHA in oil was significantly higher than that of glucose. In order to take the respective advantages of the two carbon sources, the double carbon source fermentation of glucose and glycerol was investigated, the results showed that using glucose and glycerol mixed or adding glycerol after glucose depletion were not as good as those of single carbon source, while the culture with glucose as carbon source first and supplemented with glycerol when the mass concentration of glucose in the fermentation broth decreased to 10-5 g/L could both shorten the fermentation time and increase the proportion of DHA in the oil. Using a fermentation control strategy with an initial glucose mass concentration of 20 g/L and a glycerol addition of 100 g/L during the process, the fermentation time of a 5 L bioreactor could be shortened to 56 h and the percentage of DHA in the oil could reach up to 45.50%. A control strategy for DHA production by A. limacinum using glucose and glycerol. In this paper which can significantly increase the percentage of DHA in the lipid of the cell and also effectively shorten the fermentation time, has very good industrial application value.
