Epimedium is a medicinal herb, and its main active ingredients are flavonoids, among which icariin, epimedin A, B, and C are high in content, while rare Epimedium flavonoids have high utilization value but low content. An Epimedium-flavonoid-glucosidase from Aspergillus sp. y48 strain can hydrolyze 7-O-Glc glycosyl of epimedin A and C, and convert them into sagittatoside A and C. It can also hydrolyze 7-O-Glc glycosyl and 3-O- glycosyl of epimedin B to generate icariin II and icariin. In this paper, epimedin A and C were converted into sagittatoside A and C with a purity of 98%, and the molar yields were above 95%. Using this enzyme, epimedin B was converted into sagittatoside B with a purity of 83%. The enzyme reaction time was prolonged to prepare icariin with a purity of 98% and a molar yield of 93.8%. Therefore, epimedin A, B, and C, which have high content in Epimedium, were successfully prepared into rare Epimedium flavonoids by enzymatic conversion. This method has the advantages of a simple process, high conversion rate, low cost, and high product purity. There are no other related reports until now. The results of this study provide methods for the efficient utilization of flavonoids in Epimedium and the production of rare Epimedium-flavonoids.
