Study on extraction and bioactivity of conjugated linoleic acid from Tan sheep tail fat

Abstract

In this paper, Tan sheep tail was used as raw material, low-temperature crystallization combined with urea inclusion as the method, and the yield and purity of conjugated linoleic acid (CLA) were used as evaluation indicators. The extraction process conditions were investigated, and the biological activity of Tan sheep tail fat CLA was explored. Results showed that the optimal conditions for extracting CLA from Tan sheep tail fat by low-temperature crystallization method were that the extraction solvent was absolute ethanol, the crystallization temperature was -20 ℃, the crystallization time was 12 h, and the ratio of mixed fatty acids to anhydrous ethanol was 1∶12 (g∶mL), the purity was (18.62±0.51) mg/g. On this basis, the urea inclusion method was adopted, when the inclusion temperature was 25 ℃, the inclusion time was 14 h, the mass ratio of fatty acid to urea was 1∶2, and the mass ratio of urea to ethanol was 1∶5 (g∶mL), the effect was best, and the purity was (28.23±2.36) mg/g. The results of in vitro antioxidant indicators showed that the scavenging rates of DPPH radical and ABTS cationic radicals increased with the increase of CLA concentration in Tan sheep tail fat. When the mass concentration was 6 mg/mL, the scavenging rates reached (35.94±4.07)% and (51.76±5.57)%. The results of cholesterol-lowering activity indicators showed that the inhibition rate of cholesterol micelles was (60.33±5.88)%, and the binding rates to sodium taurocholate, sodium glycocholate, and sodium cholate were (16.11±4.01)%, (37.56±3.17)%, and (25.11±4.82)%, respectively. The results provide a reference for future research on the industrial extraction and active application of Tan sheep tail fat CLA.
