Study on the preventive effect of Lactobacillus rhamnosus AFY05 on acute gouty arthritis in mice

Abstract

The objective of this study is to investigate the mechanism of action of Lactobacillus rhamnosus AFY05 in the context of acute gouty arthritis in mice.To achieve this, a mouse model of acute gouty arthritis was established using sodium urate.The effect and mechanism of AFY05 in preventing acute gouty arthritis in mice were then evaluated by assessing foot swelling, pain threshold, blood biochemical indicators, histopathological changes, and mRNA expression changes.The findings from the animal experiment revealed that AFY05 reduced foot swelling and increased pain threshold in mice.Moreover, AFY05 enhanced the enzyme activity of superoxide dismutase (SOD) and the level of glutathione (GSH) in the ankle joint tissues of mice with acute arthritis, while reducing the enzyme activity of myeloperoxidase (MPO) and the level of malondialdehyde (MDA).Additionally, AFY05 decreased the levels of interleukin-6 (IL-6), IL-10, IL-1β, and tumor necrosis factor-alpha (TNF-α) in the serum of mice with acute arthritis.Histopathological observations indicated that AFY05 alleviated tissue damage in the ankle joints of mice.Results from the qPCR experiment demonstrated that AFY05 downregulated the mRNA expressions of extracellular signal-regulated kinase 1/2 (ERK1/2), cyclooxygenase-2 (COX-2), prostaglandin E2 (PGE2), IL-6, interferon gamma (IFN-γ), and TNF-α in the ankle joint tissues of mice with acute arthritis.Furthermore, AFY05 upregulated the expressions of copper/zinc superoxide dismutase (Cu/Zn-SOD), manganese superoxide dismutase (Mn-SOD), and catalase (CAT).These findings suggest that Lactobacillus rhamnosus AFY05 exhibits potential in preventing acute gouty arthritis, surpassing the efficacy of glucosamine sulfate.Thus, it can be regarded as a strain with probiotic potential.
