Exploring the effects of flavonoid extracts from Ginkgo biloba leaves on antioxidation and anti-aging of Caenorhabditis elegans is to provide theoretical basis for the development and utilization of G. biloba leaves.The flavonoids were extracted from G. biloba leaves by reflux extraction, and the content of total flavonoids was determined by spectrophotometry.The components of flavonoid extracts were analyzed by ultra-high performance liquid chromatography tandem triple quadrupole mass spectrometry (UPLC-MS/MS).The effects of flavonoid extract of G. biloba leaves on life span, locomotion ability, reproductive ability, swallowing ability, acute heat stress ability, acute oxidative stress ability, reactive oxygen species (ROS) and lipofuscin accumulation of C.elegans were measured.The result showed that the mean yield ratio and content of total flavonoids extracted from G. biloba leaves were 8.02% and 73.68 mg/g, respectively.Totally 107 flavonoids were detected from the extract by UPLC-MS/MS.DPPH(1,1-diphenyl-2-picrylhydrazyl) free radical, ABTS (2,2′-azino-bis-3-ethylbenzthiazoline-6-sulphonic acid) cation free radical, and hydroxyl radical scavenging rates of 500 μg/mL flavonoids extract were 87.48%, 71.82% and 62.84%, respectively.The flavonoid extract of G. biloba leaves could significantly prolong the life span (P<0.001), increase the frequencies of body bending (P<0.001) and head swing, reproductive ability (P<0.001), and enhance swallowing ability (P<0.001), acute heat stress and acute oxidative stress (P<0.001), inhibit the accumulation of reactive oxygen species (ROS) and lipofuscin of C.elegans (P<0.001).The flavonoid extract of G. biloba leaves has obvious antioxidation and anti-aging effects on C.elegans and is worth of further development and utilization.
