Tryptophol (IET), a derivative of L-tryptophan (Trp), its indole structure makes it a key intermediate in synthesizing diverse pharmaceutical molecules.Currently, the industrial production of IET is entirely based on chemical synthesis, while research into its de novo biosynthesis remains limited.Because of the complex synthesis pathway of IET, metabolic engineering operations on a single-strain platform can be difficult.Furthermore, multiple genetic modifications are likely to impose a metabolic burden on the single chassis strain. These problems can be resolved by the modular co-culture strategy.By constructing an engineered Escherichia coli-Saccharomyces cerevisiae consortium, the IET biosynthetic pathway was divided into two modules. The upstream E.coli strain t12 is able to secrete Trp into the medium by overexpressing trpEDCBA, aroG and serA.To convert Trp to IET, the downstream S.cerevisiae was engineered to overexpress ARO9, ARO10, and SFA1 via genomic integration. The engineered consortium yielded 0.23 g/L IET from glucose in a 5-L bioreactor after optimizing the initial inoculation ratio and inoculum size. The methodology and findings from this study provide a conceptual framework and reference guide for the de novo biosynthesis of tryptophol and other tryptophan derivatives.
