Abstract

The mechanism underlying how the types of carbon sources influence changes in microbial community structure during the preparation of artificial pit mud using aged pit mud as a seed source remains unclear. This study investigated the effects of glucose and lactate on microbial metabolism and community structure during the restructuring process of pit mud microbiota. An anaerobic microcosm system was employed to cultivate pit mud under strictly anaerobic condition. The dynamics of substrates and metabolites were determined, and amplicon sequencing technology was used to analyze the dynamic characteristics of microbial communities. The results showed that although the relative abundance of caproate-producing bacteria in simulated fermentation decreased, they still remained capability of normal metabolism and achieved caproate content of (9.52±0.23) g/kg.Lactate was an important carbon source for pit mud bacteria to produce caproate, whereas, the addition of glucose slowed down the production rate of short- and medium-chain fatty acids. The process of new-making pit mud inevitably led to the massive growth of lactic acid bacteria.Although, the relative abundance of Methanobacteria did not exhibit a significant change, yet it substantially suppressed the relative abundances of Bacteroidia and Methanomicrobia.Based on these results, this study provides theoretical support and practical basis for further optimizing the preparation of artificial pit mud.
