Using HPLC method determine the content of β-carotene and VE in fresh pumpkin and pumpkin by freeze-drying treatment in the same time.The results showed that the calibration curve was good linear regression in the range of(1~6)μg/mL for β-carotene and(0.375~2.25)μg/mL for VE.In fresh pumpkin pulp,the average recoveries of β-carotene and VE are 100.78%(RSD=0.12%)and 99.75%(RSD=0.3%).In fresh pumpkin peel,the average recoveries of β-carotene and VE are 100.43%(RSD=0.27%)and 99.25%(RSD=0.16%)respectively.But in pumpkin by freeze-drying pulp the average recoveries of β-carotene and VE are 100.62%(RSD=0.44%)and 99.71%(RSD=0.15%)respectively.In pumpkin by freeze-drying peel,the average recoveries of β-carotene and VE are 100.11%(RSD=0.13%)and 99.75%(RSD=0.07%)respectively.The method is simple,high precision and good repeatability,and provides a reliable scientific basis for pumpkin in determining β-carotene and VE in the same time.
