Isolation and identification of lactic acid bacteria from breast milk in Xinjiang and screening of potential probiotics

Abstract

Breast milk is the main source of nutrients for infants and is rich in lactic acid bacteria. In order to understand the composition of lactic acid bacteria in breast milk in Xinjiang, bifidobacteria with excellent probiotic properties were screened for breast milk. In this paper, lactic acid bacteria in six parts of breast milk in Xinjiang were isolated and identified by traditional pure culture method. The probiotic Bifidobacterium lactis BB12 was used as a control, and Bifidobacterium was screened by fermentation characteristics, in vitro gastrointestinal fluid tolerance and bile salt tolerance tests. Two Bifidobacterium strains with potential probiotic properties were obtained. The physiological and biochemical characteristics such as optimum pH, optimum temperature, tolerance to NaCl, growth curve and acid production ability of the strains were tested. A total of 69 strains of lactic acid bacteria were isolated from 6 samples and identified as 11 species and subspecies of 7 genera, of which Streptococcus salivarius was the dominant bacteria. Two strains of Bifidobacterium with potential probiotic properties were obtained by preliminary screening: Bifidobacterium longum IMAU99727 and Bifidobacterium animalis subsp. lactis IMAU99728, which are expected to be used as new breast milk probiotics for other probiotic properties research and development and utilization.
