A method for the determination of nicotinic acid in onion by high performance liquid chromatography with diode array detector was established, and nicotinic acid in onion from different regions was studied. After ultrasonic extraction of the sample with 0.1% formic acid water, Agilent ZORBAX Eclipse Plus C18 liquid chromatographic column (250 mm×4.6 mm, 5 μm) was used, with methanol and 0.1% formic acid solution as mobile phase, a flow rate of 1.0 mL/min, column temperature of 35 ℃, injection volume of 10 μL. The detection range of the diode array detector was 190-400 nm, and the detection wavelength was 260 nm. The linear correlation coefficient R in 1-100 μg/mL of the quercetin concentration was 0.999 91-0.999 98 and the detection limit of quercetin was 0.15 mg/kg, and the quantification limit of quercetin were 0.50 mg/kg. The recovery rate of the three concentration spiking levels of 10, 20, and 30 μg/mL was 98%-103% and the relative standard deviations were 0.08%-0.11%. The effect of extraction and analysis was 1.5-2 times that of the reported methods. The method had the advantages of simple extraction, good response, good reproducibility and recovery rate, excellent precision and accuracy, and could be used for the analysis and detection of niacin in onions. It could be used for the analysis and detection of nicotinic acid in onions. The analysis of nicotinic acid in onion from 8 major production areas in China found that the content of quercetin in Jiuquan was the highest.
