摘要

以吸附量、解吸率、洗脱流速等为考察指标,确定持久霉素的最佳纯化工艺为:Ab-8型树脂具有最佳的吸附及洗脱效果,在树脂柱中静止吸附的时间为0.5h,上柱液质量浓度为13.03mg/mL,最佳上样量为20mL,洗脱流速为5mL/min,用700mL的70%溶剂A与30%溶剂B去除杂质,得到纯度较高的持久霉素,经过葡聚糖凝胶纯化后的试样中,持久霉素的相对含量可达到95%以上。

Abstract

Macroporous resin was selected for the separation of Enramycin.Ab-8 resin was the best for its highest adsorption and desorption properties.The optimum purification conditions were as follows:adsorption time 0.5 h,the concentration of adsorption liquid 13.03 mg/mL,adsorption volume 20 mL,elution flow rate 5 mL/min.700 mL 70% solvent A and 30% solvent B was used to remove impurities.After being purified by the sephadex,the purity of enramycin was up to 95%.
