地生枝顶孢深层培养富硒的研究

摘要

研究了不同浓度无机硒对地生枝顶孢RCEF0260菌株液体培养生物量、菌丝体中有机硒、多糖、甘露醇、麦角甾醇、腺苷4种主要活性成分含量的影响,并以生物量和菌丝中有机硒含量为参考指标对含无机硒的培养基进行了优化。结果显示:培养基中无机硒浓度为1.5μg/mL时RCEF0260菌丝生长状况最佳,生物量较对照提高15.99%,菌丝中有机硒含量提高908.64%,麦角甾醇含量提高较对照提高31.10%,多糖含量提高4.89%,甘露醇含量提高3.15%。培养基优化后所得RCEF0260菌丝干重为3.62 g/200 mL,较优化前提高了53.39%;菌丝中有机硒含量为93.47μg/g,较优化前提高了48.27%。

Abstract

The effects of different concentrations of inorganic selenium on biomass,the contents of organic seleni-um and 4 main functional components including polysaccharides,cordycepic acids,ergosterol and adenosine in myce-lia of Acremonium terricola in submerged culture were studied,and the Se-medium was optimized with biomass and or-ganic selenium in mycelia as criteria.Results: The mycelium of RCEF0260 grew best at 1.5 μg/mL Se,the biomass and contents of organic selenium,ergosterol,polysaccharides and mannitol were increased by 15.99% 、908.64%,31.10%,4.89% and 3.15% respectively.After optimization,the biomass in mycelia of RCEF0260 reached 3.62 g/200 mL and organic selenium content reached 93.47 μg/g,each increased by 53.39% and 48.27%.
