裙带菜酸性多糖的分离纯化与鉴定

摘要

利用水煮法及乙醇分级沉淀获得裙带菜粗多糖(UPPS-2),粗多糖经DEAE离子交换层析获得2个纯化组分(UPPS-2.1、UPPS-2.2)。对UPPS-2.1、UPPS-2.2样品进行了系列的理化特性鉴定(包括分子质量、糖含量、蛋白含量、硫酸基含量、葡萄糖醛酸含量、单糖组成及含量、糖苷键构型等)。试验结果表明:UPPS-2.1、UPPS-2.2的相对分子质量分别为1.5×105、9.4×104;糖含量分别为27.49%、31.06%;蛋白含量分别为0.00%、1.14%;硫酸基含量分别为16.90%、26.33%;二者的单糖组成相同(包括海藻糖、鼠李糖、阿拉伯糖、葡萄糖、半乳糖、木糖和甘露糖),但组成单糖的含量差异较大,前者以岩藻糖(85.78%为主,而后者主要以岩藻糖(35.93%)和甘露糖(55.76%)为主;红外光谱测定结果表明,二者均具有多糖特征吸收峰,糖苷键构型主要为β-糖苷键。经纯化后获得多糖组分UPPS-2.1是以β-糖苷键连接的主要由岩藻聚糖为主的杂多糖硫酸酯,而UPPS-2.1则是以β-糖苷键连接的主要由岩藻糖和甘露糖组成的杂多糖硫酸酯。

Abstract

To isolate and identify the acid polysaccharides from Undraria pinnatifida.Methods: Crude polysaccharides were extracted by alcohol grade precipitation from boiled Undraria pinnatifida solvent,then the crude polysaccharides were seperated by DEAE-Cellulose 52 chromatography.Two fractions obtained after elution were named UPPS-2.1 and UPPS-2.2.The physicochemical properties of UPPS-2.1 and UPPS-2.2 were studied including molecular weight,percentage of total sugar,protein,sulfate,and composition of monosaccharides,patterns of glucosidic bond.Results: The molecular weight of UPPS-2.1 and UPPS-2.2 was 1.5×105,9.4×104 respectively,the percentage of total sugar was 27.49%,31.06 %,the percentage of protein was 0.00%,1.14%,the percentage of sulfate wa 16.90%,26.33% in UPPS-2.1 and UPPS-2.2 respectively.Monosaccharide composition of UPPS-2.1 and UPPS-2.2 was same,but the percentage of monosaccharides was different by ion chromatography.The purified polysaccharides is mainly composed of β-glucosidic bonds.The main monosackcharide of the UPPS-2.1 was Fuc(85.78%),whereas in UPPS-2.2 were Fuc(35.93%) and Man/Xyl(55.76%).The SO42-of polysaccharides is link up with C2 or C3 in Fuc.Conclusion: The UPPS-2.1 was mainly composed of Fuc sulfate heterosaccharides linked by β-glucosidic bond.Whereas the UPPS-2.2 was mainly composed of Fuc and Man/Xyl sulfate heterosaccharides linked by β-glucosidic bond.
