脂肪酶非水相催化合成植物甾醇酯

摘要

研究了脂肪酶在非水相中催化合成植物甾醇酯,并利用单因素和正交实验优化酯化反应条件。结果表明:在酸醇摩尔比为3:1,溶剂添加量6 mL/g(底物),催化剂用量11%(底物百分比),反应温度45℃,反应48 h的条件下,植物甾醇的酯化率达到最大值(94.62%);产物经分离纯化后纯度达到98.2%;对产品采用薄层色谱、质谱和红外光谱进行结构表征分析,确定产物为植物甾醇酯。因此,脂肪酶可在非水相反应介质中高效催化合成植物甾醇酯。

Abstract

In the paper,the synthesis of phytosterol laurate in non-aqueous phase with lipase was studied.The optimal conditions were also obtained by single-factor and orthogonal experiments.The results showed that the yield of esterification reached the maximum(94.62%),when under the conditions of molar ratio of lauric acid to phytosterol 3∶1,adding amount of organic solvent 6mL/g,reaction temperature 45℃,lipase 11%,and reaction time 48 hrs.After purification,the content of phytesterol ester reached 98.2%.The structure of phytosterol laurate was confirmed with TLC,FT-IR and MS.Lipase can catalyze the synthesis of phytosterol esters in non-aqueous phase high efficiency.
