该研究通过液体深层发酵制备灵芝、灵芝-玉竹、灵芝-桦树茸、灵芝-人参以及灵芝-三七发酵液冻干粉,MTT细胞毒性实验及细胞形态学检测筛选出样品给药浓度,建立300 mJ/cm2中波紫外线(ultraviolet radiation b,UVB,290~320 nm)辐射人角质形成细胞(human immortalized keratinocytes,HaCaT)的损伤模型,酶联免疫吸附测定(enzyme linked immunosorbent assay,ELISA)法检测HaCaT细胞分泌炎症因子白细胞介素-1α(interleukin-1α,IL-1α)及前列腺素E2(prostaglandinE2,PGE2)的含量。以此来探究灵芝、灵芝-玉竹等发酵液冻干粉对UVB诱导的HaCaT细胞光损伤的修复作用,筛选出最优的灵芝发酵组合方式。研究结果发现,相比于灵芝发酵液冻干粉,灵芝-桦树茸发酵液冻干粉对IL-1α及PGE2的分泌有显著的抑制作用(P<0.05)。综上,相较于灵芝发酵液冻干粉,灵芝-桦树茸发酵液冻干粉修复HaCaT细胞UVB光损伤的能力最强,因此通过添加桦树茸进行灵芝发酵为最优的发酵组合方式,这为灵芝组合发酵的工业化生产提供了前提和基础。
