摘要 采用一种新的工业过程操作优化方法——可视化优化方法,对利用生物表面活性剂硫酸粘杆菌素通透性处理重组大肠杆菌BL21生产海藻糖合成酶的工艺条件进行优化。结果表明:以1.40 g/L的硫酸粘杆菌素为渗透剂,35℃渗透处理72 min,即得透性化细胞海藻糖合酶,酶活是传统超声破碎法制得的2.09倍。所得透性化细胞间歇反应时,以30%麦芽糖为底物进行海藻糖转化实验,转化率可达70%,重复使用16批(12 h/批),转化率可保持在65%以上。

Abstract: For optimizing the technological conditions of producing trehalose synthase from E. coli BL21 after permeated by colistin sulfate,a novel visualized method for the operation of industrial process called visualized optimization method was adapted in this paper. The results indicated that a proper process was achieved with cell suspension treated by 1. 40 g /L colistin sulfate for 72 min at 35℃ and the obtained enzyme activity was 2. 09 times higher than that from traditional method. Utilizing the permeabilized cells for batch fermentation,the maximum conversion rate could reach 70% with 30% maltose as the substrate. The cells could be also used for at least 16 batches( 12 h per batch),and the conversion rate kept above 65%.
