对芡实多糖的提取工艺进行了研究,并探讨了其对羟自由基的清除效果。通过单因素试验分别考察了浸提温度、浸提时间、乙醇浓度、水料比对芡实多糖提取量的影响,并确定了各因素的适用范围。利用Design-Ex-pert软件设计了正交试验,并通过响应面分析法确定了芡实多糖的最佳提取工艺条件。结果表明:芡实多糖的最佳提取工艺条件为浸提温度92℃,浸提时间6 h,乙醇体积分数85%,水料比26∶1(mL∶g),提取的芡实多糖提取量为0.62 mg/g,其对羟自由基的清除率为46.42%。
The extraction of crude polysaccharides from Semen euryales was studied and the scavenging activity of hydroxyl radical of the polysaccharides was also discussed.The effects of extraction temperature,extraction time,ethanol concentration,liquid to solid ratio on the extraction yield of the crude polysaccharides and the scavenging activity of hydroxyl radical of the polysaccharides were investigated.Single factor experiments and the orthogonal experiments was designed by Design-Expert software.The model was established by the response surface methodology analysis and the optimal extraction condition was obtained as follows: extraction temperature 92℃,extraction time 6h,ethanol concentration 85%,liquid to solid ratio 26:1,the extraction yield of crude polysaccharides was 0.63mg /g and the scavenging activity of hydroxyl radical was 46.42%.
