黏红酵母是一种可以生产类胡萝卜素的单细胞真菌,番茄红素是一种未环化类胡萝卜素,可以运用代谢控制手段,调控和阻遏某些相关的反应,使其累积番茄红素。作者基于红酵母中类胡萝卜素的合成途径,对环化抑制剂烟碱、麦角固醇合成抑制剂酮康唑、MoG还原酶激活剂青霉素、氧化剂过氧化氢等代谢调节物的调控条件进行了研究,发现添加一定量的上述代谢调控物质确实可以促进黏红酵母中番茄红素的累积。试验中采用均匀设计法优化了代谢调控的条件,研究结果表明,分别在发酵的24 h加入烟碱2.5 mL/L和青霉素4 mg/L,36h时添加双氧水1.2 mL/L,60h时添加酮康唑400 mg/L,可以使番茄红素累积量达到176.97 mg/L,是未用代谢调控时番茄红素产量4.10 mg/L的43.2倍。
Rhodotorula glutinis,a kind of caroteniod producing unicellular epiphyte,is widespread in nature.As lycopene is a carotenoid without cyclization,metabolic control means can be used to control and deter some of the related response to accumulate lycopene.On the basis of biosynthesis path of caroteniod in Rhodotorula,experiments were carried out to stduy the regulation conditions of the conditioners,such as cyclase inhibitor nicotine,ergosterol synthesis inhibitor ketoconazole,MoG reductase activator penicillin and oxidant hydrogen peroxide.We identified that lycopene was accumulated with adding this regulator above.The results revealed that the highest lycopene concentration was obtained when 2.50 ml/L nicotine,4mg/L penicillin,1.2 mL/L aquae hydrogenii dioxide(30% Hydrogen peroxide) and 400mg/L ketoconazole were added respectively at 24h,24h,36h and 60h following Rhodotorula inoculation.It reached to 176.97mg/L,43.2 times of initial lycopene yield 4.10 mg/L.
