摘要

以鱼片干为研究对象,采用高效液相色谱-荧光检测法研究酪氨酸含量与辐照杀菌处理的关系。建立了样品提取、分离和浓缩的前处理方法和液相检测方法,并提出以邻酪氨酸和对酪氨酸含量的比值作为判定样品是否辐照的标准。方法检出限为16.6~21.0 ng/mL。对酪氨酸和邻酪氨酸在16~3 800 ng/mL内均具有良好的线性关系(R2=0.999 7~0.999 9),样品加标回收率(n=3)分别为82.7%~98.3%和83.3%~93.8%,相对标准偏差(RSD)<7.8%。

Abstract

This paper investigated the relationship between tyrosin composition and irradiation sterilized dried fish.The tyrosin composition was determined by high performance liquid chromatogram-fluorescence detector.The method of sample preparation was also developed,including extraction,separation and concentration.The ratio of p-tyr and o-tyr was used to identify whether sample was irradiated.The detection limit was 16.6~21.0 ng/mL.The linear range was from 16 ng/mL to 3 800 ng/mL,and linear correlation was good(R2 = 0.999 7~0.999 9).Recovery of o-tyrosine and p-tyrosine were 82.7% ~98.3% and 83.3 %~93.8%,respectively.The relative standard deviation was less than 7.8%.
