Constipation, a common gastrointestinal disorder, significantly impacts the life quality of patients and disrupts their daily activities.Studies have shown that constipation is associated with gut microbiota dysbiosis.Previous animal studies have demonstrated that Lacticaseibacillus rhamnosus CCFM1068 alleviates constipation.Building on this, this study aimed to evaluate the effect of L.rhamnosus CCFM1068 on constipation and its role in improving the gut microbiota in constipated individuals through clinical trials. In this study, 25 patients with constipation were recruited from Wuxi Second People’s Hospital. After a one-week screening period, eligible patients underwent a four-week probiotic intervention.Fecal samples were collected from each patient before and after the intervention to analyze changes in gut microbiota.Concurrently, spontaneous bowel movements (SBM), the bristol stool form scale (BSFS), the patient assessment of constipation symptoms (PAC-SYM), and the quality of life (PAC-QOL) were assessed weekly. Results indicated that after four weeks of L.rhamnosus CCFM1068 intervention, the frequency of defecation, stool consistency, and life quality of patients were significantly improved.Furthermore, the overall composition of the gut microbiota in patients underwent significant changes. At the phylum level, Firmicutes abundance was significantly increased, while Bacteroidetes abundance was decreased. At the genus level, L.rhamnosus CCFM1068 improved stool consistency and alleviated constipation by downregulating the abundance of Bacteroides, Phocaeicola, Alistipes, Parabacteroides, Odoribacter, and Butyricimonas, while upregulating that of Dorea, Clostridium, Anaerobutyricum, Raoultella, Adlercreutzia, Megamonas, Mediterraneibacter, Streptococcus, and Escherichia. These findings provide a theoretical foundation for the future application of probiotics in alleviating constipation.
