该文通过向柴达木双层环伞菌发酵体系中添加黑果枸杞花色苷粗提物,研究其对菌体生长和胞外多糖(exopoly saccharides,EPS)合成的影响,优化了花色苷的添加方式和发酵温度工艺,同时对调控体系中的残糖(还原糖)含量、pH及溶解氧的动态变化进行研究。研究表明,相较于空白对照组,采用半流加黑果枸杞花色苷粗提物(发酵0~36 h流加,速率0.83 mL/min,总添加量1 800 mL),变温培养(0~36 h为23 ℃,36~96 h为25 ℃,96~120 h为23 ℃)方式,菌丝生物量提高89.04%、多糖产量增加65.46%。从构建的菌体生长、EPS产量及底物消耗的动力学模型可知,调控组模型拟合曲线的相关系数均>0.93,拟合情况良好,说明模型能很好地反映发酵过程规律。因此,黑果枸杞花色苷能够显著促进柴达木双层环伞菌生长与EPS的合成。该研究为青藏高原特色食用菌的规模化开发与应用提供理论支持。
