Abstract

Milk fat globule membrane protein (MFGMP) is an important functional component of the milk fat globule membrane (MFGM).Its unique structure and biological properties play multiple key roles in infant health and development. This study employed an in vitro static model to simulate the infant gastrointestinal digestion of human and bovine MFGM and its proteins, and monitored dynamic changes in free amino acids during digestion.Peptidomics analysis revealed the differences in the composition of polypeptides. The mouse macrophage RAW264.7 model assessed the anti-inflammatory activity of digestive products from different sources. The results show that human MFGMP digestion products contain significantly higher levels of both essential (Thr, Val, Ile, Leu, Lys) and non-essential (Gly, Ala, Ser, Glu, Asp) amino acids than bovine sources, and contain six unique bioactive peptides. All four digestive products show no cytotoxicity and have certain proliferation-promoting effects on cells at 200 and 400 μg/mL.Measurements of nitric oxide (NO) and immune factors indicate that human MFGMP digestive products possess superior anti-inflammatory potential. Moreover, the MFGM structure shows no significant impact on this anti-inflammatory effect.
