茶籽壳原花青素柱分离条件的研究

摘要

通过静态吸附与解吸的柱分离实验,筛选出大孔吸附树脂HPD200A对茶籽壳原花青素具有较好的分离效果;通过动态吸附与解吸的柱分离实验,考察了上样流速、pH值、洗脱流速、洗脱液pH值、洗脱液浓度等因素对大孔吸附树脂HPD200A吸附及解吸花青素(PC)的影响。结果表明,动态条件下的检测指标,随考察因子的梯度变化而有符合理论规律的变化。综合生产效益及影响因素,柱分离条件采用上样液pH值5.0、上样流速1.50mL/min上样;用pH值7.0的体积分数20%和60%乙醇水溶液以0.50mL/min的流速分步洗脱,并得到纯度分别为97.22%和76.46%的原花青素物质。

Abstract

In the paper,the macroporous resin HPD200A ,which was considered to have better effects of separation for procyanidins from episperm of tea seeds,were selected by method of adsorption and desorption in static state. The effects of separated procyanidins by HPD200A materials were studied according to factors which influenced the PC absorption and desorption in dynamic experiments. The loading speed of sample,value of pH,eluted speed,eluted fluid pH and concentration of ethanol were tested. The result showed that the detection index under the dynamic condition fitted for the theoretical changes. Considering the influence factors and yield ratio,the best condition of separation was:sample loading speed:1.50 ml/min (pH 5.0); eluted sequentially by 20 % ethanol (pH 7.0) and 60 % ethanol (pH 7.0) at speed of 0.50mL/min,purity were 97.22 % and 76.46 % respectively.
