摘要

通过使用添加不同浓度的葡萄糖和果糖的YNB培养基,使酵母在高浓度糖的环境中生长,并通过与低浓度糖培养相比较发现:酵母在高浓度糖培养环境中,Crabtree效应严重,自身生理代谢发生重大的改变,有氧呼吸被强烈的抑制,造成TCA循环断裂,从而导致发酵初期柠檬酸、α-酮戊二酸和苹果酸大量地分泌到胞外,乳酸的发酵副产物含量增大,但当糖浓度降低时,TCA循环重新闭合,柠檬酸、α-酮戊二酸和苹果酸又重新进入细胞参与代谢。

Abstract

In the paper,YNB culture with high concentration of glucose and fructose was used for the growth of Saccraomyces cerevsiae. It showed that huge changes of metabolism in Saccraomyces cerevsiae had occurred,such as the Crabtree effect,the inhibition of aerobic respiration and the breakage of the TCA cycle,which caused the secretion of citric acid,α-ketoglutarate and malate out of the cell and the increase of the by-product concentration like lactic acid at the beginning of fermentation. Then,the sugar was consumed gradually as the process went on,which led to the second TCA cycle,and also made citric acid,α-ketoglutarate and malate entering into the cell consequently. To summarize,the mechanism of Saccraomyces cerevsiae cultured under high concentration sugar was profiled,and could serve as the theoretical support to beer manufacturers.
