菊芋可溶性蛋白的提取及其分子质量的测定

摘要

利用磷酸缓冲液提取菊芋可溶性蛋白,采用单因素实验和正交实验研究提取工艺条件,确定的提取条件为:料液比(w/v)1∶40、处理时间4h、处理温度40℃、pH值8.0。在此条件下,蛋白质平均提取率为14.7mg/g。采用十二烷基硫酸钠-聚丙烯酰胺凝胶电泳(SDS-PAGE)测定所提菊芋蛋白质的分子质量,结果显示其分子质量介于20.9-69.2ku之间。

Abstract

The soluble protein of Jerusalem artichoke was extracted by the phosphate buffer. Single experiment and orthogonal experiment were used to discuss the optimized extraction,and the SDS-PAGE was used to analyze furthermore. The results showed that the optimal extraction conditions are as follows:ratio of material to solution 1:40(w/v),extraction time 4h,temperature 35℃,pH8.0. Under this condition,the average extracting rate of soluble protein was 14.7mg/g. In the SDS-PAGE,the molecular weights of these proteins were between 20.9 ku and 69.2 ku.
